چکیده

With the use of stigmatic exudate or distilled water as carriers, various antimetabolites, inhibitors, and miscellaneous materials were injected into the hollow styles of detached Lilium longiflorum pistils before, at, or after compatible or incompatible pollination. Pollen tube lengths were measured 48 hr after pollination with pollinated styles incubated at 22-23 degrees C. Substances considered inhibitors of protein synthesis in microbial systems significantly retarded both compatible and incompatible pollen tube growth while inhibitors of RNA synthesis tended to significantly inhibit compatible pollen tube growth with less or no effect on incompatible pollen tubes. Application of the inhibitors in stigmatic exudate at or after compatible pollination produced significant results at the lowest concentrations. Significant retardation of pollen tube growth also occurred after injection of 2,4-dinitrophenol, mercaptoethanol, indoleacetic acid, naphthaleneacetic acid, benzyladenine, dimethyl sulfoxide, or potassium or sodium iodide. Pollen tube growth in detached pistils of L. longiflorum may be useful as a bioassay in situ for screening biologically active materials.

Incorporation of label into pollen tube walls from myoinositol-labeled Lilium longiflorum pistils.

Compatible and incompatible pollen tubes growing on detached Lilium longiflorum pistils which had been prelabeled with myoinositol-U-(14)C take up a portion of the label and utilize it for biosynthesis of tube wall substance. The label is transferred from pistil to pollen tubes apparently via the secretion products (exudate) of the pistil. The exudate thus appears to have a major nutritional ro...

Correlation between the Accumulation of Stylar Canal Exudate and the Diminution of Self-incompatibility in Aging Flowers of Lilium longiflorum

Self-incompatibility in Lilium longiflorum cv. Hinomoto diminishes with aging of the flower. The relation between the diminished self-incompatibility of the aged flower and stylar canal exudate in Lilium longiflorum was studied. Carbohydrate content in stylar canal exudate increased with aging of the flower, indicating that it accumulated with time. During the aging process, the molecular weigh...

The Nutritional Role of Pistil Exudate in Pollen Tube Wall Formation in Lilium longiflorum: II. Production and Utilization of Exudate from Stigma and Stylar Canal.

Detached pistils of Lilium longiflorum were labeled with d-glucose-U-(14)C 24 hours after anthesis and then sampled for the next 6 days to determine the appearance of label into exudate from the stylar canal and the stigmatic surface of the pistil. Results were obtained with unpollinated cv. Ace pistils and pollinated cv. Ace pistils, selfed or crossed (cv. Croft pollen). Limited data were also...

Autoradiographs of Pollen Tube Nuclei with Calcium-45

Autoradiography with Ca(45) has been used to obtain information about the relation between calcium and chromosomes. Labelled pollen from the Easter lily, Lilium longiflorum, was allowed to develop into pollen tubes between 5 and 6 cm. long in the styles of non-radioactive flowers. All of the nuclei, namely the tube nucleus and the two sperm nuclei, retain Ca(45) after this period of growth and ...

pH gradients are not associated with tip growth in pollen tubes of Lilium longiflorum.

The cytoplasmic pH of growing pollen tubes of Lilium longiflorum Thunb. was measured using the pH-sensitive fluorescent dye 2',7'-bis-(carboxyethyl)-5(6')-carboxyfl uorescein and confocal fluorescence ratio imaging. The average cytoplasmic pH in the clear zone of the pollen tube tip was pH 7.11, and no consistent pH gradients were detected in the clear zone, averaging around -1.00 milli pH unit...
